The SP5X uses a Koheras white light laser source in addition to regular short wavelength lasers
The Leica SP5X confocal system is equipped with multiple laser sources, including a 405 diode, an Argon laser (458, 476, 488, 496, 514nm) and a white light laser. The white light laser emits a broad spectrum of wavelengths from 470 to 670nm in 1 nm increments for simultaneous optimal excitation of multiple fluorophores.

Tunable excitation and tunable detection
Allow the user to image samples that have been labeled with any fluorophore. Excitation scans allow optimization of excitation wavelength for new fluors, for combinations of fluors, or to effectively avoid autofluorescence artifacts.

Tunable Prism-Based Spectral Detector and GaAsP hybrid detector
These allow the user to choose between detectors; the resonant (HyD) detector allows imaging with minimum laser power to minimize photo-toxicity and bleaching.

High precision x-y and Z-Galvo motorized scanning stage
The microscope has a high precision z-galvo Z stage that has a minimum step size of 3nm and a travel range of 1500 µm. This facilitates high-resolution 3D reconstruction.

Live-cell imaging with environmental chamber
The microscope has a transparent environmental chamber that enables users to prolong studies of living cells, including time-lapse image capture experiments for cell motility and cell death. Warm, filtered air circulates within the acrylic chamber from a heater unit that is mechanically isolated from the environmental chamber. Two top doors allow specimens to be changed. Another two doors below the stage allow access to the microscope setting controls.
